School of Science Division of Life Science 34 Genetic Studies on Stem Cell Regulation Supervisor: XIE Ting / LIFS Co-supervisor: TU Renjun / LIFS Student: LEE Man Ping / BIOT Course: UROP1100, Fall UROP2100, Spring UROP3100, Summer Kinases take an important role in the phosphorylation processes in vivo in order to maintain normal cell functioning. Some cyclin-dependent kinases are being identified to be involved in the self-renewal or differentiation of germline stem cells in Drosophila. However, the function of many kinases in Drosophila remains unknown. Here, I have conducted a genetic screening for 50 transgenic RNAi genes encoding for kinases to determine their roles in GSC self-renewal and differentiation. 23 genes out of the 50 genes investigated show a reduction in the number of germline stem cells, in which the knockdown of CG17528, cerk, cdk9 and CG11811 show more significant phenotypes. In future studies, I will further investigate the interaction mechanisms of the kinases identified in the germline stem cell niche. Utilizing stem cells could cure severe diseases. Stem cell therapy such as introducing genetically modified hematopoietic stem cells into a patient’s bone marrow could cure leukemia. Induced pluripotent stem cells are found to turn an adult cell back into a stem cell, increasing the flexibility of the usage of stem cells. Drosophila serves as a common model for scientists to discover the mechanisms within a different types of stem cells. That knowledge may be useful in understanding certain similar genes related to stem cells in humans. Therefore, this report includes a summary of stem cell research genetic strategies in Drosophila.
