In response to its local microenvironment, macrophages can polarize into M1 phenotype which has pro-inflammatory characteristic, and M2 phenotype which perform anti-inflammatory feature. However, macrophage polarization disorder can damage tissue during inflammatory diseases and promote development of tumor. Hence, regulating macrophage polarization in specific disease could be a promising therapeutic strategy. Various peptide sequences are known to be responsible in macrophage polarization. These peptide signals were conjugated on the surface of micro-sized hydrogel and were presented as solid bound signals. The effect of peptide bound signal and its peptide combination on macrophage polarization were examined. M1 macrophages promotes inflammatory response and anti-microbial activities while M2 group macrophages serves as pro-healing and anti-inflammatory activator. (Jayasingam, S. et al., 2020; Orecchioni, M. et al., 2019; Gordon, S., 2017) Modulators including peptides on hydrogel can induce diverse polarization. However, the exact mechanism behind M1/M2 polarization and the effect of external factors including the morphology of the hydrogel is still unknown. To understand and predict this complex relationship for immunomodulation, hydrogel presenting peptides with different combinations are generated and different machine learning algorithms are built to understand its complex relationship between different properties of peptide, hydrogel and macrophage polarization.
