School of Science Division of Life Science 18 Receptor Based Drug Development from Chinese Herbal Medicine Supervisor: LI, Ning / LIFS Co-supervisor: XUE, Hong / LIFS Student: LIU, Zhenru / BCB Course: UROP1100, Spring Traditional Chinese medicine (TCM) has long employed the medicinal plant Scutellariae Radix (SR), sometimes referred to as Baical Skullcap Root. Numerous pharmacological properties of SR have been discovered, including anti-inflammatory, anti-cancer, antioxidant, and neuroprotective actions. The active ingredients of SR incorporate flavonoids, baicalin, baicalein, wogonin, and oroxylin A, which can pass through the blood-brain barrier and perform their biochemical functions. Multiple components contribute to diverse and even opposite effects of SR as a common feature of herbal medicine. In this report, the anxiolytic effects and potential side effects of depression and hyperactivity are evaluated in mouse model under administration of SR extracts. Mice were subjected to behavior analysis after being tested in elevated plus maze, tail suspension and locomotor experiments, providing hint to further studies of SR and its therapeutic application. Brain tumors come in a wide variety of forms. While some brain tumors are benign, others are malignant. Cancer may start in other body regions and move to the brain as secondary brain tumors, or brain tumors may start in the brain themselves. Brain tumor growth rates might vary significantly. How a brain tumor impacts the nervous system’s work depends on its development rate and location. This semester, we have done cancer cell cultures, DNA library constructions, chromatography of Chinese Herbal powders, and living cell imaging to set up the foundations of future research on Chinese Herbal Medicine for brain cancer treatment. From previous studies, gene mutation is the main cause of cancer, while those changes might represent a possible target for the treatment of cancer, which makes the identification of the SNPs in the DNA sequence of cancerous cells important for further investigation of treatments for cancers. In this project, Inter-Alu PCR is used as a tool for library preparation before undergoing next-generation sequencing by using primers of Alu elements, which are transposable elements widely distributed in human genes, facilitating the genome-wide sequencing of cancerous cell DNA and determination of the whole genome of the cell.
